The drug affinity complex is a maleimidopropionic acid group attached to a lysine side chain. It reacts with the free thiol of cysteine-34 on circulating albumin, forming a covalent bond. This conjugation keeps the peptide in the bloodstream and shields it from rapid renal filtration and proteolysis. Reported circulation half-lives for the albumin-bound form fall in the range of roughly six to nine days in early human studies.
Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
CJC-1295 belongs to a class of synthetic peptides modeled on growth hormone releasing hormone, a forty-four amino acid signal produced by the hypothalamus. The compound is built from the first twenty-nine residues of the natural sequence, with four substitutions introduced at positions 2, 8, 15 and 27. These changes were designed to slow enzymatic breakdown while preserving receptor activation. The result is a peptide that is shorter than native GHRH and considerably more resistant to ordinary clearance pathways in circulation.

Two related forms circulate in technical discussion under the same family name. The original version carries a drug affinity complex (DAC) that binds covalently to serum albumin after administration, and this linkage substantially extends circulation time. A second form, frequently written as modified GRF(1-29) or CJC-1295 without DAC, lacks that linker and clears much faster. The naming is a frequent source of confusion because the shorthand CJC-1295 can refer to either form depending on the source. Reports sometimes fail to specify which variant was studied.
The peptide backbone includes a D-alanine at position two, which resists cleavage by dipeptidyl peptidase IV, and several other substitutions that reduce degradation. Its molecular weight is roughly 3.4 kDa without the linker and about 3.6 kDa with it. The molecule is water soluble and is normally supplied as a lyophilized powder. Precise sequence and mass values depend on which variant is described, so technical documents usually state the exact form being referenced.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.
The albumin-binding version stays in circulation for days, because covalent attachment to serum albumin shields the peptide from rapid filtration and degradation. Reported half-lives for this form fall in the range of several days. The version without the linker is cleared in minutes, with estimates often near thirty minutes in animal work. These figures come from small studies and vary with assay method, species, and route, so they are best read as approximate rather than fixed constants.
Studies in this area generally track growth hormone pulses, insulin-like growth factor 1 concentrations, and occasionally body composition endpoints. Most published human data come from early, small trials, and questions about long-term effects remain open. Whether repeated exposure alters pituitary responsiveness over time is not settled. Analytical work relies on immunoassays for the hormones and on mass spectrometry for the peptide itself, because the two measurements answer different questions.
== Economy == Bophuthatswana was the richest of the TBVC-states as it had platinum mines, which accounted for two-thirds of the total platinum production in the Western world. It was also rich in asbestos, granite, vanadium, chromium and manganese. Additional revenues came from the Sun City casino, which was a day trip from Johannesburg and Pretoria, where gambling was illegal under the National Party government, as it was throughout all of South Africa. Bophuthatswana had a relatively capitalist free market economy, as seen by the government's drive for infrastructure development, foreign direct investments and virtually no regulatory barriers to starting and running a business. Initially, its economy was driven by agriculture then later by mining, hospitality and banking. Bophuthatswana also issued bearer development bonds. The so-called "Bop Bonds" were redeemed by the government of the North West province from 1995 to 1997, and are now worthless as financial instruments. However, bonds in excellent condition are considered collectible. Bonds issued in 1988 and 1989, in R10 and R20 denominations, currently trade at 10–25% of original face value.
== Medical complications and limitations == The Chinese study Breast Augmentation by Autologous Fat-injection Grafting: Management and Clinical analysis of Complications (2009) reported a reduced incidence of medical complications with strict control of the rate of injection (cm3/min) of the volume of breast-filler by injecting the fat-grafts in even layers within the breast-tissue matrix. The small (2-mm.) incision and blunt-cannula injections reduce the possibility of damaging the underlying structures of the breast (milk ducts, blood vessels, nerves). Injected fat-tissue grafts that do not establish an adequate blood supply can undergo necrosis from lack of oxygen and result in oil cysts that will become calcified. The medical complications (sclerotic lesions, and breast disease) occurred to the 17-patient group were identified and located with X-ray computed tomography and MRI visualizations of the breast tissues. The sclerotic lesion was excised and the liquefied fat was evacuated; the excised samples indicated biological changes in the intramammary fat grafts, such as fat necrosis, fat calcification, fat hyalinization, and fibroplasia.
Calcium is released from the sarcoplasmic reticulum into the sarcomere when a muscle is stimulated to contract. This calcium uncovers the actin-binding sites. When the muscle no longer needs to contract, the calcium ions are pumped from the sarcomere and back into storage in the sarcoplasmic reticulum. There are over 600 skeletal muscles in the human body (exact number varies by counting method) and skeletal muscle makes up ~40% of human mass.
A split range pressure controller may also modulate a Joule-Thomson valve across the turbo-expander. Pressure in blanketed tanks is maintained by self actuating pressure control valves (PCVs). As liquid is withdrawn from the tank the pressure in the gas space falls. The blanket gas supply valve opens to maintain the pressure. As the tank fills with liquid the pressure rises and a vent gas valve open to vent gas to atmosphere or a vent system. Rupture (bursting) discs (PSE) and pressure relief or pressure safety valves (PSV) are important pressure control devices. Both are self-actuating and are designed to open at a preset pressure to provide an essential safety function on the petrochemical plant.

=== Development of databases === HMRBase: A manually curated database of hormones and their receptors. It is a compilation of sequence data after extensive manual literature search and from publicly available databases. HMRBase can be searched on the basis of a variety of data types. Owing to the high impact of endocrine research in the biomedical sciences, HMRBase could become a leading data portal for researchers. The salient features of HMRBase are hormone-receptor pair-related information, mapping of peptide stretches on the protein sequences of hormones and receptors, Pfam domain annotations, categorical browsing options, and online data submission. This database is integrated with DrugPedia so the public can contribute. BIAdb: A database for Benzylisoquinoline Alkaloids. The Benzylisoquinoline Alkaloid Database serves to gather information related to the BIA's. Many BIA's show therapeutic properties and can be considered as potent drug candidates. This database will also serve researchers working in the field of synthetic biology, as developing medicinally important alkaloids using synthetic process is one of the important challenges. This database is also integrated with DrugPedia so the public can contribute. Antigen DB: This database contain more than 500 antigens collected from literature and other immunological resources. These antigens come from 44 pathogenic species. In Antigen DB, a database entry contains information regarding the sequence, structure, origin, etc.
This drastically revamped King Crimson had no plans to record in the studio, focusing instead on playing "reconfigured" versions of past material live. According to Levin, the band "were instructed/advised by Robert Fripp to look at the older classic King Crimson material as if we had written it. And so we did that with a lot of older material that the band had done before the '80s. We didn't actually cover that much of the '80s material outside of a few songs." For the most part, this approach would remain consistent for the remainder of the band's lifetime. In early 2014, the band's repertoire included songs from the run of albums between In the Court of the Crimson King and Larks' Tongues in Aspic for the first time since 1974, as well as reviving song material from Red. No songs with vocals by Belew were included in the setlist, although some instrumentals from his tenure were played. Some material from A Scarcity of Miracles was also incorporated into the band's repertoire. After rehearsing in England, King Crimson toured North America from 9 September to 6 October. Recordings from the Los Angeles dates were released as Live at the Orpheum; this included the newly-written instrumentals "Banshee Legs Bell Hassle" and "Walk On: Monk Morph Chamber Music". Tours of Europe, Canada, and Japan followed in the later half of 2015. Brand-new songs mainly written by Fripp and Jakszyk were debuted at the concerts, as well as drum showcases.
Iranians have one of the highest per capita rates of tea consumption in the world. Châikhânes (teahouses) are common in Iran. Iranian tea is typically served in traditional Iranian glasses with a traditional saucer and teaspoon. Tea is cultivated in northern Iran along the shores of the Caspian Sea. In Burma (Myanmar), tea is consumed not only as hot drinks, but also as sweet tea and green tea known locally as laphet-yay and laphet-yay-gyan, respectively. Pickled tea leaves, known locally as lahpet, are a national delicacy. Pickled tea is usually eaten with roasted sesame seeds, crispy fried beans, roasted peanuts and fried garlic chips. In Mali, gunpowder tea is served in series of three, starting with the highest oxidisation or strongest, unsweetened tea, locally referred to as "strong like death", followed by a second serving, where the same tea leaves are boiled again with some sugar added ("pleasant as life"), and a third one, where the same tea leaves are boiled for the third time with yet more sugar added ("sweet as love"). Green tea is the central ingredient of a distinctly Malian custom, the "grin", an informal social gathering that cuts across social and economic lines, starting in front of family compound gates in the afternoons and extending late into the night, and is widely consumed in Bamako and other large urban areas. In the United States, 80% of tea is consumed as iced tea. Sweet tea is native to the southeastern U.S. and is iconic in its cuisine due to its refreshing temperature and large amount of sweetener.
In contrast to the elemental ions sodium, potassium, and calcium, there is no known cellular mechanism specifically dedicated to regulating intracellular lithium. Lithium can enter cells through epithelial sodium channels. Lithium ions interfere with ion transport processes (see "Sodium pump") that relay and amplify messages carried to the cells of the brain. Mania is associated with irregular increases in protein kinase C (PKC) activity within the brain. Lithium carbonate and sodium valproate, another drug conventionally used to treat the disorder, act in the brain by inhibiting PKC's activity and help to produce other compounds that also inhibit the PKC. Lithium carbonate's mood-controlling properties are not fully understood.

== Advocacy == Ammann spent much of his life fighting injustices within the health care system, nationally and internationally. From 1992 to 1996 he served as director of research programs for the Pediatric AIDS Foundation. Ammann was also the chairman of the Conference on Global Strategies for Prevention of HIV Transmission from Mothers to Infants. Ammann became a member of the American Foundation for AIDS Research (AmFAR) Board of directors in 1988, and was also appointed chairman of the AmFAR scientific advisory committee. From 1997 to 1998 Ammann served as president of AmFAR. In these positions, Ammann fought for an increase in research funding in HIV, in particular for women and children, and the development of more cost-effective treatments. In 1997, Ammann founded Global Strategies for HIV Prevention. In his position as president, Ammann advocated for the needs of the most marginalized populations that are affected by the HIV epidemic. The organization is aimed at helping to prevent HIV transmission from mothers to infants in order to slow the spread of the HIV virus.
=== Central nervous system stimulants === Some systematic reviews and meta-analyses of clinical research using low doses of certain central nervous system stimulants have indicated that these drugs may enhance cognition in healthy people. In particular, the classes of stimulants that demonstrate possible cognition-enhancing effects in humans have evidence in vitro as direct agonists or indirect agonists of dopamine receptor D1 or adrenoceptor α2. Relatively high doses of stimulants cause cognitive deficits.
=== Mutation === IDH1 mutations are heterozygous, typically involving an amino acid substitution in the active site of the enzyme in codon 132. These mutations are somatic, meaning they primarily occur in cells that can become cancerous, such as those in brain and bone tumors. The mutation results in a loss of normal enzymatic function and the abnormal production of 2-hydroxyglutarate (2-HG). It has been considered to take place due to a change in the binding site of the enzyme. 2-HG has been found to inhibit enzymatic function of many alpha-ketoglutarate dependent dioxygenases, including histone and DNA demethylases, causing widespread changes in histone and DNA methylation and potentially promoting tumorigenesis.
=== The Finke era with ten Bundesliga seasons (1991–2007) === SC Freiburg were promoted to the 2. Bundesliga in 1978–79, which they would compete in for a decade-and-a-half before making the breakthrough to the top-flight Bundesliga in 1993–94 under the management of Volker Finke. In their first Bundesliga season, Freiburg narrowly avoided relegation. They made a third-place finish in their second season at the top level, just three points behind champions Borussia Dortmund. It was at this time that they were first nicknamed Breisgau-Brasilianer (literally Breisgau-Brazilians), due to their attractive style of play. The club reached the UEFA Cup in 1995, where they were knocked out in the first round by Slavia Prague. In 2001 they reached the UEFA Cup for a second time, where they were knocked out by Feyenoord. Freiburg's first Bundesliga relegation was in 1997 after they finished in 17th position. While they have been relegated four times since first making the Bundesliga, they have thrice won immediate promotion back to the top league. It was the first time since 1992 that Freiburg played in the 2. Bundesliga for two consecutive seasons. Freiburg finished the 2006–07 season in fourth place in the 2. Bundesliga, missing out on the third automatic-promotion spot on goal difference to MSV Duisburg, although they won 12 of their last 16 league games. They were knocked out of the DFB-Pokal in the second round by VfL Wolfsburg on 24 October 2006. On 20 May 2007, Volker Finke resigned as the club's coach after 16 years in the job.
Not only did the people of ancient Mexico use the same methods of production as the Phoenicians, they also valued murex-dyed cloth above all others, as it appeared in codices as the attire of nobility. "Nuttall noted that the Mexican murex-dyed cloth bore a "disagreeable ... strong fishy smell, which appears to be as lasting as the colour itself." Likewise, the ancient Egyptian Papyrus of Anastasi laments: "The hands of the dyer reek like rotting fish". So pervasive was this stench that the Talmud specifically granted women the right to divorce any husband who became a dyer after marriage. In 2021, archaeologists found surviving wool fibers dyed with royal purple in the Timna Valley in Israel. The find, which was dated to c. 1000 BC, constituted the first direct evidence of fabric dyed with the pigment from antiquity.

The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
It has not been approved by major regulatory agencies for any therapeutic indication. Material sold under this name is typically distributed for laboratory research only. Clinical use is not supported by large controlled trials.
Early human studies of the albumin-binding form reported circulation half-lives on the order of several days. The value depends on assay method and study design. Estimates for the form without the binding moiety are considerably shorter.
The names are often used interchangeably in casual writing, but they are not strictly identical. Modified GRF(1-29) refers to the unconjugated analogue carrying only the four substitutions. The version sold as CJC-1295 with DAC includes the albumin binding linker and clears much more slowly.
