CJC-1295 is a synthetic peptide designed to mimic growth hormone-releasing hormone (GHRH), the endogenous signal that prompts the pituitary gland to release growth hormone. The compound is a modified fragment of the natural hormone, spanning the first twenty-nine amino acids of GHRH with several substitutions that slow enzymatic breakdown. Two variants circulate in research settings: one carrying a drug affinity complex (DAC) and one without it. The DAC-free form is frequently labelled Mod GRF(1-29) in catalogs and discussion forums.
The peptide binds GHRH receptors on somatotroph cells within the anterior pituitary, triggering a signalling cascade that increases growth hormone secretion. Its improved resistance to dipeptidyl peptidase IV degradation distinguishes it from the parent hormone. In the DAC-bearing version, a maleimide group reacts with a cysteine residue on serum albumin, forming a covalent bond that keeps the peptide in circulation far longer. That albumin attachment is the central design feature separating the two research variants.
Pharmacokinetic behaviour differs sharply between the two forms. The DAC-bearing peptide shows an extended circulation time measured in days, whereas the version without the complex is cleared within roughly half an hour. This gap shapes how researchers design dosing schedules in animal models. Whether the prolonged presence of the DAC form produces effects meaningfully different from the short-acting variant remains an open question, since comparative human data are scarce.
Lyophilized peptide powder is comparatively stable when kept dry, cold, and protected from light. Once dissolved, the molecule is vulnerable to deamidation, oxidation, and aggregation, with the rate depending on pH, buffer composition, and temperature. Alkaline conditions and repeated freeze-thaw cycles accelerate loss of the intact peptide. The methionine present in the native sequence is a known oxidation site, which is one reason it was replaced in the modified fragment. Suppliers typically recommend cold storage of solutions and use within a short window.
Analytical confirmation usually relies on reversed-phase high-performance liquid chromatography for purity and on liquid chromatography coupled to mass spectrometry for identity. Mass data reveal the expected molecular mass and can flag truncated or oxidized species. Amino acid analysis and peptide mapping provide sequence-level verification. Immunoassays are used in some biological matrices, but antibodies raised against one releasing-hormone analog may cross-react with another. Reported purity figures depend heavily on the method used, so comparisons between suppliers require matching the analytical approach.
The two variants differ dramatically in how long they persist in circulation. The form lacking the albumin-binding group has a plasma half-life measured in tens of minutes, comparable to the natural hormone fragment. The version carrying the drug affinity complex binds albumin and shows a half-life of roughly six to eight days in human studies. That figure comes from small trials that tracked hormone levels over extended periods. The physiological consequences of sustained versus pulsatile stimulation are still debated and the literature does not settle the point.

Lyophilized material is typically stored at minus twenty degrees Celsius or lower. Keeping the vial dry and protected from light preserves peptide integrity. Repeated freeze-thaw cycles can cause aggregation or loss of activity. Once dissolved, solutions are generally kept at two to eight degrees Celsius. Stability data for reconstituted solutions vary, and long-term behavior is not fully established. Working aliquots reduce the number of times a stock container is opened.
Reverse-phase high-performance liquid chromatography is the standard tool for purity assessment. The technique separates the target peptide from truncated or modified byproducts. Mass spectrometry confirms molecular weight and supports sequence verification. Electrospray ionization and matrix-assisted laser desorption are both used. Amino acid analysis provides an independent check on composition. Purity values are commonly reported as area percentage from the chromatogram. Residual trifluoroacetate and water content are also measured in many quality programs.
Batch-to-batch consistency depends on solid-phase peptide synthesis and subsequent purification. Coupling efficiency, resin choice, and cleavage conditions all affect the final profile. Counter-ion content and moisture can shift the apparent mass of a batch. Documentation typically includes a certificate of analysis with chromatograms and spectra. Independent verification by a second laboratory is sometimes requested. Whether a given certificate reflects the actual vial contents depends on chain of custody. Analytical methods themselves carry uncertainty that should be stated alongside results.
The compound emerged from work at a Canadian biotechnology firm in the early 2000s. Early human studies examined its effect on growth hormone and insulin-like growth factor 1 in healthy volunteers and in people with HIV-associated fat redistribution. Reports described sustained increases in both markers after a single injection. Development did not advance to regulatory approval, and the clinical programme was later discontinued. The molecule is now encountered mainly as a research chemical rather than a marketed medicine.
Two forms circulate in research settings and are frequently confused. One carries the drug affinity complex and is often written as CJC-1295 with DAC; the other lacks that group and is usually called modified GRF(1-29). The two share the same core sequence but differ sharply in how long they persist in blood. Products labelled only as CJC-1295 normally refer to the version carrying the complex. Documentation that omits the distinction leaves the intended molecule ambiguous.
CJC-1295 is a synthetic peptide built as a long-acting analogue of growth hormone-releasing hormone. Its backbone matches the first twenty-nine residues of the natural human hormone, with four amino acid substitutions added to slow enzymatic breakdown. A reactive maleimide group, commonly termed the drug affinity complex, allows the peptide to attach to circulating albumin after administration. That albumin attachment keeps the molecule in the bloodstream for an extended period instead of being cleared within minutes.
In 1981, a commercial involving a train was made, the musical theme was "La Colegiala" composed by Rodolfo Aicardi. Due to the enormous popularity of Nescafé, during the Second World War, "all the production of the American plant was reserved only for the use of the military". In Ecuador, a Boeing 737-200 from the AeroGal company was painted red to promote the brand. In Chile, since 2009 the brand has sponsored and helped to restore a well-known Chilean theater that was in decline, making it the first Nescafé theater in the world and naming it the Nescafé Theater of the Arts. In previous years, the brand was sponsoring different stars of Canal 13, such as Esta Noche Fiesta and Tuesday 13, the 123 Nescafé competition and was for some time the sponsor of different campaigns of the Chilean Telethon, returning as sponsor to the campaign in 2011. In turn, its variant Nescafé Dolca was sponsor of Una Vez Más of Canal 13. In the Philippines, an advertising commercial was released in 2020 with their newest jingle and slogan, "Babangon tayo, susulong tayo" ("We will rise, we will advance"). English rock band Muse successfully sued Nescafé in 2003 when their song "Feeling Good" was used in a television ad without permission and donated the £500,000 compensation to Oxfam.
== Contraindications == Apomorphine is contraindicated when used concomitantly with serotonin 5-HT3 receptor antagonists, including ondansetron, granisetron, and palonosetron, as this combination has been associated with profound hypotension and loss of consciousness. It is also contraindicated with allergies to the drug or its excipients, including sulfites. Intravenous administration of apomorphine is associated with blood clots and pulmonary embolism due to crystallization in the vein and is not recommended.
== Publication == In July 2016, Hillbilly Elegy was popularized by an interview with Vance in The American Conservative. The volume of requests briefly disabled the website. Halfway through August, The New York Times wrote that the title had remained in the top ten Amazon bestsellers since the interview's publication. Vance has credited his Yale contract law professor Amy Chua as the "authorial godmother" of the book, as she persuaded him to write the memoir.
Polysulfated glycosaminoglycan (PSGAG; sold under the brand name Adequan) is an injectable drug for dogs and horses that is used to alleviate the limpness, pain, and lowered range of motion caused by arthritis. It is made of repeat disaccharide units (comprising hexosamine and hexuronic acid), and is similar to glycosaminoglycans already present in the cartilage; PSGAG thus easily integrates itself there. In vitro studies have shown it to inhibit the enzymes that degrade cartilage and bone, as well as suppress inflammation and stimulate the synthesis of replacement cartilage. While it can cause an increased risk of bleeding, it is relatively safe and has a high LD50. PSGAG is one of the most widely prescribed joint treatments for horses. While it is widely used, some studies still show conflicting results in terms of efficacy, causing some to claim that PSGAG is not solely responsible for the significant mitigation of arthritis seen in success cases.

== Biochemistry == PCT is a member of the calcitonin (CT) superfamily of peptides. It is a peptide of 116 amino acids with an approximate molecular weight of 14.5 kDa, and its structure can be divided into three sections (see Figure 1): amino terminus (represented by the ball and stick model in Figure 1), immature calcitonin (shown in Figure 1 from PDB as the crystal structure of procalcitonin is not yet available), and calcitonin carboxyl-terminus peptide 1. Under normal physiological conditions, active CT is produced and secreted in the C-cells of the thyroid gland after proteolytic cleavage of PCT, meaning, in a healthy individual, that PCT levels in circulation are very low (<.05 ng/mL). The pathway for production of PCT under normal and inflammatory conditions are shown in Figure 2. During inflammation, LPS, microbial toxin, and inflammatory mediators, such as IL-6 or TNF-α, induce the CALC-1 gene in adipocytes, but PCT never gets cleaved to produce CT. In a healthy individual, PCT in endocrine cells is produced by CALC-1 by elevated calcium levels, glucocorticoids, CGRP, glucagon, or gastrin, and is cleaved to form CT, which is released to the blood. PCT is located on the CALC-1 gene on chromosome 11. Bacterial infections induce a universal increase in the CALC-1 gene expression and a release of PCT (>1 μg/mL). Expression of this hormone occurs in a site specific manner. In healthy and non-infected individuals, transcription of PCT only occurs in neuroendocrine tissue, except for the C cells in the thyroid.
carriers were in a vulnerable position, and the low visibility coupled with radar confusion let a Japanese bomber slip through and severely damage USS Franklin. Although the American carrier aircraft strikes caused less destruction to enemy naval vessels than earlier battles, American submarines made up for it by sinking two of the three Japanese fleet carriers, which left Zuikaku as the only remaining operational IJN fleet carrier. The American F6F Hellcat fighter proved its worth, as its powerful engine generated superior speed, while its heavier armor and firepower made it rugged and deadly. The Japanese on the other hand were still flying the A6M Zero which, though highly maneuverable and revolutionary during the early stages of the Pacific War, was now underpowered, fragile and essentially obsolete by 1944. In addition, the D4Y "Judy", though fast, was also fragile and easily set on fire. Japanese naval airmen were also inadequately trained. The Japanese training programs could not replace the quality aviators lost during the past two years of the Pacific Campaign. Flying against the well-trained and often veteran U.S. aviators, it was a one-sided contest. The Americans lost fewer than two dozen Hellcats in air-to-air combat. Naval aviation and anti-aircraft fire shot down nearly 480 Japanese aircraft, 346 of those carrier aircraft on 19 June alone.
Computer simulations indicated energy barriers of 0.61–0.75 eV for hydroxyl-terminated atomic defects that participate in a Grotthuss-type relay, while pyrylium-like ether terminations did not. Recently, Paul and co-workers at IISER Bhopal demonstrated solid state proton conduction for oxygen functionalized few-layer graphene (8.7 × 10−3 S/cm) with a low activation barrier (0.25 eV).

When a drug is developed with evidence throughout its history of research to show it is safe and effective for the intended use in the United States, the company can file an application – the New Drug Application (NDA) – to have the drug commercialized and available for clinical application. NDA status enables the FDA to examine all submitted data on the drug to reach a decision on whether to approve or not approve the drug candidate based on its safety, specificity of effect, and efficacy of doses.

Dihydroergotamine (DHE), sold under the brand names D.H.E. 45 and Migranal among others, is an ergot alkaloid used to treat migraines. It is a derivative of ergotamine. It is administered as a nasal spray or injection and has an efficacy similar to that of sumatriptan. Nausea is a common side effect. It has similar actions to the triptans, acting as an agonist to the serotonin receptors and causing vasoconstriction of the intracranial blood vessels, but also interacts centrally with dopamine and adrenergic receptors. It can be used to treat acute intractable headache or withdrawal from analgesics.

The synthetic peptide keeps the receptor-binding region of GHRH but carries substitutions that resist enzymatic cleavage. In the DAC version, an added group also anchors the molecule to albumin, extending its presence in the bloodstream.
The maleimide group forms a covalent link with albumin, an abundant blood protein. This attachment slows clearance, so a single administration persists much longer than the unmodified peptide. The feature is the main reason the two variants are handled differently in study design.
It is not an approved therapeutic product in major regulatory jurisdictions. It appears in research chemical catalogs and scientific literature rather than pharmacy shelves. Clinical development was limited and did not reach approval.
The version carrying the affinity complex bears a maleimide group that binds serum albumin, which extends its circulation time to several days. The version without it lacks this group and clears within roughly half an hour. The two are chemically related but behave very differently once in the body.
