Binding to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and stimulates growth hormone synthesis and release. Because the peptide acts upstream of the pituitary, effects are mediated through endogenous growth hormone rather than direct receptor activation in peripheral tissues. Increases in insulin-like growth factor 1 are generally described as a downstream consequence. Most published human exposure data come from small early-stage studies, and the clinical significance of the pharmacokinetic profile remains incompletely characterized.
CJC-1295 is a synthetic analogue of growth hormone-releasing hormone, built on the 29-amino-acid GHRH(1-29) fragment. Four substitutions distinguish it from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes reduce enzymatic cleavage and extend the peptide's persistence in circulation. The compound is discussed in two forms, one carrying a drug affinity complex and one without it.
At the pituitary, the peptide binds the growth hormone-releasing hormone receptor on somatotroph cells. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone. Somatostatin and other hypothalamic signals modulate this response. Negative feedback from insulin-like growth factor 1 also influences output. The same regulatory architecture operates with the native hormone. Whether the synthetic analog alters feedback dynamics over repeated exposure remains an open question. Most published receptor work uses cell models rather than intact human systems.
CJC-1295 is a synthetic peptide belonging to the growth hormone-releasing hormone analog family. It comprises twenty-nine amino acid residues derived from the N-terminal region of natural GHRH. The molecule incorporates several non-natural substitutions that increase resistance to enzymatic degradation. These modifications extend its activity compared with the native hormone fragment. Researchers use it to study pituitary growth hormone secretion in laboratory and clinical settings. This compound is distinct from native GHRH in its stability profile.
Two principal forms appear in the literature and in research supply. One carries a drug affinity complex, a maleimide-based group that forms a covalent bond with circulating albumin. This linkage slows clearance and produces a long-lasting elevation of peptide levels. The other form lacks that group and is often labeled MOD GRF(1-29). It has a much shorter circulation time. Both variants retain the same core receptor-binding sequence. Reported half-lives differ substantially between the two.

The attached maleimide group explains the unusual duration of the DAC version. After injection it reacts with the thiol of cysteine-34 on serum albumin, forming a stable covalent bond. The resulting conjugate is too large for rapid kidney filtration and is shielded from many peptidases. Reported half-lives for this form reach several days, whereas the version without the group is cleared in roughly half an hour. That gap is the main pharmacological difference between the two.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.
Both forms act at the pituitary receptor for growth hormone-releasing hormone and increase growth hormone output, which in turn raises insulin-like growth factor 1. A long-acting analog produces sustained rather than pulsatile stimulation, and the physiological consequences of that pattern are not fully settled. Published human data on the extended form remain limited, and much of what circulates in discussion traces to early company reports rather than independent replication. How sustained exposure affects normal feedback remains an open question.
The distinguishing feature of the DAC form is a maleimide-containing group that reacts with the free thiol of cysteine-34 on human serum albumin. This reaction forms a covalent bond without enzymatic assistance, and it takes place after the peptide enters the bloodstream. Because albumin is abundant and long-lived, the attached peptide is carried through circulation far longer than an unmodified fragment would survive. The chemistry is a deliberate pharmacokinetic strategy rather than a change to receptor activity.
Receptor level activity follows the canonical GHRH pathway. The peptide binds the GHRH receptor, a class B G protein coupled receptor on somatotroph cells of the anterior pituitary. Binding raises intracellular cyclic AMP, which promotes calcium entry and the release of growth hormone into the bloodstream. Because the molecule acts at the same receptor as the endogenous hormone, its effect is superimposed on the natural pulsatile rhythm rather than replacing it. Whether sustained receptor occupation leads to desensitization is not fully settled.
CJC-1295 belongs to a class of synthetic peptides modeled on growth hormone releasing hormone, a forty-four amino acid signal produced by the hypothalamus. The compound is built from the first twenty-nine residues of the natural sequence, with four substitutions introduced at positions 2, 8, 15 and 27. These changes were designed to slow enzymatic breakdown while preserving receptor activation. The result is a peptide that is shorter than native GHRH and considerably more resistant to ordinary clearance pathways in circulation.
The amended article now defines the participation of the Federal Council and the 16 German states in matters concerning the European Union. Article 29 was again modified and provided an option for the states to "revise the division of their existing territory or parts of their territory by agreement without regard to the provisions of paragraphs (2) through (7)". Article 118a was introduced into the Basic Law and provided the possibility for Berlin and Brandenburg to merge "without regard to the provisions of Article 29, by agreement between the two Länder with the participation of their inhabitants who are entitled to vote". A state treaty between Berlin and Brandenburg was approved in both parliaments with the necessary two-thirds majority, but in a popular referendum of 5 May 1996, about 63% voted against the merger. The German states can conclude treaties with foreign countries in matters within their own sphere of competence and with the consent of the federal government (Article 32 of the Basic Law). Typical treaties relate to cultural relationships and economic affairs. Some states call themselves a "free state" (Freistaat). It is merely a historic synonym for "republic" and was a description used by most German states after the abolition of monarchy after World War I. Today, Freistaat is associated emotionally with a more independent status, especially in Bavaria. However, it has no legal significance. All sixteen states are represented at the federal level in the Bundesrat (Federal Council), where their voting power depends on the size of their population.
Pakistan drew around 6.6 million foreign tourists in 2018. Pakistan boasts attractions from mangroves in the south to Himalayan hill stations in the northeast, including ancient Buddhist ruins of Takht-i-Bahi and Taxila, the 5,000-year-old Indus Valley civilisation sites such as Mohenjo-daro and Harappa, and numerous mountain peaks over 7,000 metres (23,000 feet). The northern part of Pakistan boasts numerous old fortresses, showcasing ancient architecture. It encompasses the Hunza and Chitral valleys, where the small pre-Islamic Kalasha community resides, claiming descent from Alexander the Great. Lahore, Pakistan's cultural capital, contains lots of Mughal architecture, including the Badshahi Masjid, the Shalimar Gardens, the Tomb of Jahangir, and the Lahore Fort. Following the 2005 Kashmir earthquake, The Guardian highlighted "The top five tourist sites in Pakistan" to boost tourism, featuring destinations like Taxila, Lahore, the Karakoram Highway, Karimabad, and Lake Saiful Muluk. Festivals and government initiatives aim to promote Pakistan's cultural heritage. In 2015, the World Economic Forum ranked Pakistan 125th out of 141 countries in its Travel & Tourism Competitiveness Report.
== Side effects == The most common adverse effects in studies were upper respiratory tract infection, headache, nausea, and diarrhea. The most common side effects in clinical trials were nasopharyngitis, nausea, headaches, herpes simplex (including oral herpes, ophthalmic herpes, herpes dermatitis and genital herpes), and increase in blood creatine phosphokinase. Abrocitinib can cause serious infections, malignancy, major cardiac events, thrombosis and other laboratory abnormalities including thrombocytopenia, lymphopenia, and lipid elevations. However, according to clinical data, Abrocitinib is well tolerated. The total adverse reactions were not statistically different between the placebo and the dose of 100 mg of abrocitinib. However, it was slightly higher for the dose of 200 mg of abrocitinib. Symptoms such as acne, headache, and nausea, appeared in the first two weeks of starting abrocitinib, and it was not necessary to interrupt the treatment. In general, the AE frequency of abrocitinib was the same or a little bit higher than in case of placebo or dupilumab.
From then on, the two teams interacted regularly and Watt coined the term ionosphere to describe the multiple atmospheric layers they discovered. In 1927 the two radio labs, at the Met Office and NPL, were combined to form the Radio Research Station (with the same acronym, RRS), run by the NPL with Watt as the Superintendent. This provided Watt with direct contact to the research community, as well as the chief signals officers of the British Army, Royal Navy and Royal Air Force. Watt became a well-known expert in the field of radio technology. This began a long period where Watt agitated for the NPL to take a more active role in technology development, as opposed to its pure research role. Watt was particularly interested in the use of radio for long-range aircraft navigation, but the NPL management at Teddington was not receptive and these proposals went nowhere.
=== Baked goods and confectioneries === Cakes are considered to be intermediate moisture foods because of their moisture content (18-28%), and have low enough water activity that preserve the safety and quality. Some examples of baked goods and confectionery that come under this category are fruit cakes, pie fillings, candies, marshmallows, jams, pizza crust. Tutti Fruiti is a candy-like product that can be made from a variety of fruit, most commonly papaya. Raw pieces of unripe papaya are boiled and layered with sucrose until reaching 68 degrees brix. The solution is then air dried until a moisture content of 25.7% is reached.

== Other hormones secreted == C-peptide, which is secreted into the bloodstream in equimolar quantities to insulin. It helps to prevent neuropathy and other vascular deterioration related symptoms of diabetes mellitus. A practitioner would measure the levels of C-peptide to obtain an estimate for the viable beta cell mass. Amylin, also known as islet amyloid polypeptide (IAPP). The function of amylin is to slow the rate of glucose entering the bloodstream. Amylin can be described as a synergistic partner to insulin, where insulin regulates long term food intake and amylin regulates short term food intake.
Most teleost species are oviparous, having external fertilisation with both eggs and sperm being released into the water for fertilisation. Internal fertilisation occurs in 500 to 600 species of teleosts but is more typical for Chondrichthyes and many tetrapods. This involves the male inseminating the female with an intromittent organ. Fewer than one in a million of externally fertilised eggs survives to develop into a mature fish, but there is a much better chance of survival among the offspring of members of about a dozen families which are viviparous. In these, the eggs are fertilised internally and retained in the female during development. Some of these species, like the live-bearing aquarium fish in the family Poeciliidae, are ovoviviparous; each egg has a yolk sac which nourishes the developing embryo, and when this is exhausted, the egg hatches and the larva is expelled into the water column. Other species, like the splitfins in the family Goodeidae, are fully viviparous, with the developing embryo nurtured from the maternal blood supply via a placenta-like structure that develops in the uterus. Oophagy is practised by a few species, such as Nomorhamphus ebrardtii; the mother lays unfertilised eggs on which the developing larvae feed in the uterus, and intrauterine cannibalism has been reported in some halfbeaks. There are two major reproductive strategies of teleosts; semelparity and iteroparity. In the former, an individual breeds once after reaching maturity and then dies.
In 2017, Ice-T signed a petition to Congress for criminal-justice reform. His 1994 book, The Ice Opinion, was largely focused on his view on politics. Writing in the aftermath of the 1992 Los Angeles riots, he was one of the only rappers who criticized the targeting of Koreans by some Black rioters. On June 5, 2008, Ice-T joked that he would be voting for John McCain in the 2008 American elections, speculating that his past affiliation with Body Count could hurt Barack Obama's chances if he endorsed him, so he would choose instead to ruin McCain's campaign by saying he supported him. In 2015, Ice-T said that he admired Obama "just for the simple fact he took the shot and went all the way to be president" and included Obama in the list of people who he would like to have for a dream dinner party. On September 22, 2022, Ice-T narrated an advertisement for the Foundation for Individual Rights and Expression.
"During the first three days there was continuous torture. I was beaten with an aluminum rod and with cables. ? Then I was told to sign a statement with my hands tied behind my back, so I didn't even see the paper and I don't know what I signed."

The two share the same 29-amino-acid backbone. CJC-1295 carries an additional albumin-binding moiety that markedly extends its residence time in circulation, while the form without that moiety clears faster. Literature sometimes applies the name loosely to either variant.
It has not been approved by major regulatory agencies for any therapeutic indication. Material sold under this name is typically distributed for laboratory research only. Clinical use is not supported by large controlled trials.
Early human studies of the albumin-binding form reported circulation half-lives on the order of several days. The value depends on assay method and study design. Estimates for the form without the binding moiety are considerably shorter.
It is a synthetic peptide modeled on growth hormone-releasing hormone. The molecule is used in research on pituitary growth hormone secretion. It differs from the natural hormone through several stabilizing substitutions.
